I've only got about 1/8" of clearance between one spot of the bellhousing portion of the 4L80E case, and my trans. tunnel, on the drivers side.

Any opinions on whether or not I need to make an adjustment to increase this clearance to keep it from hitting? I'm concerned about damage to the transmission when things start twisting.

I'm using standard height solid body bushings, and poly motor and trans mounts.

My torque goals for this build are 550 to 600.

It has been so long since I had this car running, I forget which side goes up and which goes down, when it starts to twist, but if I recall it is the drivers side which goes up. It might be time for me to cut off some of the housing material, or dimple the tunnel a tad.

I cut the ear off my 4L80E. Did it in the car also without damage to the bedliner coating. Used a sawzall and put a piece of thin metal between the ear and the trans tunnel so there was no damage done to the coating on the tunnel.

Jody's right - let the sawzall go to town.

Only problem with cutting the ear off, you just rendered your core price of that case to zero. Chances are I would do the same, lol.
