I disconnected my battery while my truck is winter stored. Come spring what should the start up process be? Hook up the battery and wait a while before starting?

May depend on which PCM you are using. If I recall correctly, Andrewb70 told me the Holley Term X does not use volatile memory and a bat disconnect has no impact on it.

Nope just start it up and drive it assuming GM ECM.
