I have a Spectra EFI tank I'm installing and I don't know what this fitting is:

It came with this adapter, but they don't really expect me to use a barbed fitting do they?

It's not -6 or -8.

Looks like a return for a return fuel system.

I think it is an SAE 45 degree fitting.

3/8" you can get an -AN adapter, should be a 3/8 to -6.

They make automotive double 45 degree fittings for high and low pressure, your adapter being the low pressure variety so I'm guessing your male fitting also is. Don't work much with 45 stuff so I was just commenting on size, and you mentioned -AN sizes -6 or -8 I thought you were just looking for a way to adapt the two to run braided line. What fuel system you running.

I want to plug hose with a -6 AN fitting into the 45 degree flare coming off the tank.

As noted above, that appears to be an SAE 45*.
If you decide to use the barbed fitting with a push-on hose/clamp be sure to use a high-pressure fuel injection rated hose.
