Tubing is 4", with 1 60 degree bend & 2 90s.

There is no rule that I have ever heard of. Each bend will contribute to flow loss. But with 4" diameter tubing, losses will be very small, depending on how much motor you have.
If you are less than 800 horsepower, I would not worry about the design you have shown.

OK thanks. This design I had, i discovered wont work. Coolant fitting is in the way. Going to have to get a different waterpump with fitting to the passenger side of throttle body, or move battery to driver's side, or trunk.

I have read that less than 20" cost HP but never understood how that could be. Mine has the same bends you are looking to do with 3 90s and works just fine. https://static1.pt-content.com/images/pt/2014/03/725139_zpse0f694bb-1.jpg (http://s1114.photobucket.com/user/charchri4/media/Stuff%20to%20save%20for%20web/725139_zpse0f694bb.jpg.html)
